Son and former partner of Philip Protheroe (1747-1803) in Protheroes and Claxton, MP for Bristol 1812-1820, brother of Philip Protheroe (d. 1846) (q.v.). Originally followed his father into partnership as a West India merchant and then invested in coal mines and in smelting in the Forest of Dean, buying assets for 20,000 guineas from his uncle John Protheroe in 1812.
1841 census Edward Protheroe coalmaster living at Newnham Gloucs., aged 67 with wife Anne aged 70. 1851 census Edward Protheroe aged 77 born Bristol proprietor of iron and coal mines living at 8 Eccleston Square with wife Anne aged 80.

Letters of administration of the estate of Edward Protheroe formerly of Hill House Newnham Gloucs., but late of Eccleston Square, who died at Brighton 24/08/1856, and left unadministered by Mark Davis Protheroe the nephew, granted 26/09/1870 to Lucy Hungerford Protheroe. Effects under £20.
